The Lexus GX460 is body-on-frame construction, more off-road oriented; The Mercedes ML350 and BMW X5 are unibody construction, optimized for city/highway driving. GX460 has a very good AWD system which is capable of serious off-road usage, and it comes as standard. For ML350 and X5 35i, AWD is optional, and they do not have true off-road ability.

For the same price range, the GX460 is the only one that offers a V8 engine. However its acceleration performance is slower than the ML350 and X5 due to heavier weight.

The GX460 has the 3rd-row seat which is "usable"; The ML350 only has 2 rows of seats; the BMW X5's optional 3rd-row seat is too cramp to fit an adult there (children only). So if you need a practical 3rd-row seat, the GX460 is your only choice.
